黄豆苷元对H2O2诱导的PC12细胞损伤的保护作用

摘要:目的 探讨黄豆苷元(daidizein)对H2O2诱导的PC12细胞损伤的保护作用。方法 用H2O2处理体外培养的PC12细胞建立细胞损伤模型,并加入daidizein预处理作为保护,显微镜下观察PC12细胞形态改变,MTT法检测活细胞数,LDH法检测细胞膜的损伤情况。结果 H2O2处理细胞24 h后,细胞形态发生明显改变,胞体变小、突起缩回,MTT吸光值减小,活细胞数减少,LDH释放量明显增加,与正常对组组比较差异有显著性(P<0.01)。daidizein处理组细胞形态明显改善,MTT吸光值显著增加,LDH释放量明显降低,与H2O2处理组相比差异有显著性(P<0.01)。结论 daidizein能显著抑制H2O2对PC12细胞的毒性,具有较强的神经保护作用。关键词: PC12细胞; H2O2 ;黄豆苷元
The protective effects of Daidizein on the PC12 cells injury induced by H2O2

Abstract: Objective To investigate the effect of Daidizein on the PC12 cell injury induced by H2O2. Methods PC12 cells injury model was made with H2O2 treating, and the treatment group was processed with Daidizein. The cellular morphology was examined with microscopy, the cells proliferation was measured by MTT assays, and the cell membrane injury was measured by LDH assays. Results The PC12 cellular morphology was altered with H2O2 treating for 24 h, MTT value decreased and LDH leakage increased obviously. What is different compared with control group (P<0.01). In the group pretreated with Daidizein for 2h, the cellular morphology recovered, MTT value was increased, and LDH leakage decreased, significantly different compared with H2O2 group (P<0.01). Conclusion Daidizein can inhibit the cytotoxic effect induced by H2O2 and has strong protective function on nerve cell. ......
