诊断性超声对仔鼠学习记忆及海马组织 乙酰胆碱酯酶活性的影响

     [摘要]目的 研究一定功率的超声波对仔鼠学习记忆及海马组织乙酰胆碱酯酶活性的影响,探讨诊断超声的安全性。方法 妊娠ICR小鼠40只,随机分为辐照0min组(对照组)、辐照10min组、辐照20min组、辐照30min组4组。妊娠第14天应用一定功率的连续超声波(3.5MHz,输出功率1W)对各组孕鼠进行辐照,待仔鼠生长至30d和90d时,从各组仔鼠中随机选出20只进行旷场行为实验,20只进行Y迷宫分辨学习实验。实验完毕,立即处死仔鼠,取出不同组仔鼠海马组织,测其中的乙酰胆碱酯酶活性。结果 与对照组相比,辐照20min组和30min组小鼠学习能力明显损伤(均P<0.01);辐照20min组和30min组小鼠脑内乙酰胆碱酯酶活性明显升高(均P<0.01)。结论 孕14d的母鼠接受一定剂量的超声辐照后对仔鼠的学习能力有损伤,这种损伤可能与脑内乙酰胆碱酯酶活性升高有一定的相关性。

    Influence of diagnostic ultrasound on learning and memory of neonatal

    mice and the contents of acetylcholinesterase in the hippocampus JIA Meiqun, JIANG Xiaoqing

    Abstract:Objective To investigate the influence of continuous ultrasound on mice learning and memory, and the contents of acetylchalinesterase (ACHE) in hippocampus, and to evaluate the safety of diagnostic ultrasound. Methods Forty pregnant ICR mice were randomly divided into four groups:0min group(control group),10min group,20min group,30min group. The mice were received continuous ultrasound (3.5MHz, intensity1W) on 14th day of gestation. In each group neonatal mice were randomly selected to behavioral tests (twenty for open field test and twenty for Ytype maze)for learning and memory at thirty days and three months of age. In each group mice were sacrificed after tests and activities of ACHE in hippocampus were measured. Results Compared with control group, the learning ability decreased in 20min group and 30min group(all P<0.01); the activities of ACHE in hippocampus was significantly higher in 20min group and 30min group(all P<0.01). Conclusions Exposure to some dose ultrasound of mice on 14th day of gestation may result in disturbances in fetal mouse brain.This may be related with the the activities of ACHE in mice’s hippocampus. ......
