摘 要 目的:研究血卟啉衍生物协同毫米波辐射对白血病细胞株的细胞结构、细胞凋亡率及氧化自由基的影响;方法:电镜显微技术研究细胞结构的变化,Annexin V/PI双染法结合流式细胞术分析细胞株凋亡和坏死,噻唑蓝 (MTT)法测定细胞活力;结果:血卟啉协同毫米波辐射使白血病细胞株细胞结构产生显著的退行性变化,显著诱导细胞凋亡和坏死,抑制细胞活力;结论:血卟啉对部分功率密度的毫米波辐射有增敏作用,显著促进K562细胞坏死(P<0.01),抑制细胞活力。其机制与辐射后氧化自由基水平上升及抗氧化酶系活性下降有关。关键词 血卟啉衍生物;毫米波;射频辐射;增敏;白血病
Effects of Millimeter Wave Radiation Combined with HPD on Leukemia Cell Line K562

Abstract Objective :To study the effects of millimeter wave radiation combined with HPD on cytoarchitecture,cell apoptosis rate,oxyradical and antioxidase of leukemia cell strain K562. Methods: K562 cells were immersed in HPD (10μg/mL) and radiated with millimeter wave. Then the cytoarchitectures were observed under electron microscope; flow cytometre and Annexin V/PI were used to analyze cell apoptosis and necroses; intracellular malondialdehyde (MDA) was analyzed with thiobarbituric acid method,and superoxide dismutase (SOD) was analyzed with pyrogallic acid method. Results:Irradiated by millimeter wave,remarkable increase of the content of MDA within K562 cells was observed,and the content of SOD was inhibited evidently,retrograde changes of the cytoarchitecture in K562 cells were observed under electron microscope,the increases of cell apoptosis rate and necroses rate were also observed after the irradiation. Conclusion :High power density millimeter wave irradiation combined with HPD induces significant changes of ultrastructure and remarkable increase of apoptosis and necroses rates of K562 cells. ......
