摘要:目的 克隆家蝇Ace基因并进行表达,并对家蝇AChE序列进行分析,为利用基因工程进行酶改造提供必要的参考。 方法 结合已有的文献资料,利用生物信息学方法对家蝇AChE的序列进行分析,包括密码子偏爱性、系统发生、三维结构预测等。 结果 成功克隆家蝇Ace基因,并利用同源模建方法获得了家蝇AChE的三维结构,以果蝇AChE为参考,判断了家蝇AChE应属于AChE2家族。 结论 根据所得家蝇AChE的三维模型从理论上分析了因AChE中一个氨基酸突变导致家蝇产生有机磷抗性的可能原因,并为半理性改造家蝇乙酰胆碱酯酶提供了三维模型。关键词:乙酰胆碱酯酶;序列分析;进化树;三维结构;同源模建
Cloning and sequence analysis of AChE gene from Housefly(Musca domestica).

Abstract:Objective To clone acetylcholinesterase(AChE)gene from Housefly(Musca domestica)in order to provide neces-sary reference for genetic engineering study. Methods The sequence of AchE was,by means of bioinformatics,analyzed in-cluding codon bias,systemic genesis,predicting of three-dimentional structure by using bioinformatics.Results AChE gene from housefly was cloned and AChE three-dimentional structure was obtained by homologous modeling.AChE,belonged to AChE2fami-ly,from fruitfly was taken as reference. Conclusion Based on housefly's AChE three-dimentional structure it is theoretically in-dicating the possibility that a mutation of an amino acid in Housefly's AChE can lead to the resistance of housefly to organic phospho-rus pesticide. ......
