[摘要]目的：动态观察N_苄基氟哌啶醇氯化物(F3)对大鼠胸主动脉平滑肌细胞内Ca2+的影响。方法：用Ca2+荧光染料Fluo_3_AM负载大鼠胸主动脉平滑肌细胞，在激光扫描共聚焦显微镜上测定细胞内游离钙的变化。结果：在含CaCl2(126mmol/L)的细胞外液中，30mmol/LKCl使细胞内钙荧光强度(FI)迅速增强，F3可以拮抗KCl诱导钙FI增强作用，并呈量效依赖和时间依赖关系。4种浓度F3(001、01、10、10 μmol/L)作用3min时，使KCl诱导细胞内钙FI从(81±4)%分别降至(71±18)%(n=20，P<005)、(51±12)%(n=20，P<001)、(39±14)%(n=20，P<001)、(28±14)%(n=20，P<001)。钙FI变化最快的时程是在给药后的0～30s。结论：F3通过阻断细胞膜电压依赖性钙通道降低平滑肌细胞内钙浓度，这可能是其扩张血管，改善心肌缺血的机制。[关键词]氟哌啶醇；血管平滑肌细胞；钙；激光扫描共聚焦显微镜
Effects of N_benzyl Haloperidol Chloride on Intercellular Calcium in Rat Vascular Smooth Muscle Cells

[Abstract]Objective: To investigate the effects of N_benzyl haloperidol chloride(F3)on intracellular calcium([Ca2+]i)in vascular smooth muscle cells(VSMCs)isolated from thoracic aortas of rat. Methods: VSMCs were loaded with a calcium sensitive fluorescent dye Fluo_3_AM, and [Ca2+]i was determined by the use of laser scanning confocal microscope. Results: In the extracellular solution with Ca2+ at the concentration of 126 mmol/L, 30 mmol/L KCl induced a rapid increase of intracellular calcium fluorescence intensity(FI). F3 antagonized the increase of FI induced by KCl in both concentration dependent manner and time dependent manner. After 3 minutes of the administration of F3 at the concentration of 001, 01, 10, 10 μmol/L, the increased FI of the VSMCs induced by KCl was decreased from(81±4)% to(71±18)%(n=20, P<005) ......
