【Abstract】 AIM: To study the toxicity of different doses of MnCl2 on rat pheochromocytoma PC12 cells in vitro and their effects on signal transduction of extracellular signalregulated kinase 1/2 (ERK1/2). METHODS: PC12 cell cultures were exposed to MnCl2 within the range of 100900 μmol/L. The inhibition of the cells during 7 d was determined by MTT assay and plate clone formation test, and ERK1/2 and pERK1/2 were determined by Western blot. RESULTS: MnCl2 at different concentrations (100, 300, 500, 700 and 900 μmol/L) could inhibit the growth and proliferation of PC12 cells in a dose and timedependent manner and the cell growth curves were changed accordingly (P<0.05). Western blot showed that MnCl2 decreased pERK1/2 expression in a dosedependent manner(P<0.05). CONCLUSION: MnCl2 can significantly inhibit the growth and proliferation of PC12 cells, possibly by downregulating pERK1/2 expression.【Keywords】 MnCl2; rats; pheochromocytoma PC12 cell; pERK1/2
