摘要:目的 研究白细胞介素2(IL2)和白细胞介素18(IL18)诱导自然杀伤(NK)细胞活化和抗骨髓瘤细胞活性的协同作用。方法 流式细胞仪方法检测NK细胞表面CD69的表达;阴性细胞纯化法用于获得高纯度的NK细胞; ELISA测定NK细胞培养上清液中干扰素(IFNγ)的浓度;标准铬51释放试验评估NK细胞杀伤骨髓瘤细胞的能力。结果 与单一应用IL18相比,低浓度IL2和IL18联合应用显著提高了NK细胞的活性、分泌干扰素(IFNγ)的水平和杀伤骨髓瘤细胞的作用。结论 低浓度IL2具有协同IL18活化NK细胞抗骨髓瘤细胞的作用,此结果为IL2和IL18联合应用治疗骨髓瘤奠定了一定的理论基础。关键词:白细胞介素2;白细胞介素18;自然杀伤细胞;骨髓瘤细胞
The effect of interleukin2 (IL2) synergized with IL18 to enhance activities of natural killer cells

ABSTRACT: Objective To explore potential synergetic effects of interleukin2 (IL2) with IL18 on induction of natural killer (NK) cell activity against myeloma cells in vitro. Methods CD69 expression on NK cells were defined by Flow Cytometry; IFN γ level in supernatants of NK cell culture was measured by ELISA; pure NK cell population was obtained from peripheral blood by using cell purification kit; standard Cr51 release method was used to test NK cell cytotoxicity against myeloma cells in vitro. Results The combined application of IL18 with low dose of IL2 (10u/mL) significantly enhanced NK cell activation, IFNγ secretion and NK cell cytolytic activity against myeloma cells (U266 and RPMI8226) as compared to IL18 alone, respectively. Conclusion IL2 synergized with IL18 to induce NK cells activates antimyeloma cell in vitro and combined use of IL18 and low dose IL2 could be an alterative immunotherapy approach for cancers including myeloma. ......
