穿孔素生物学活性测定法的建立及对肺癌转移预报作用的评价

摘要 目的 建立PBMC穿孔素活性的测定方法并探讨其对肺癌转移预报的价值。方法 以PBMC为效应细胞、兔红细胞为靶细胞，根据Hb的过氧化物酶样作用，以OPD为显色底物测定靶细胞裂解后释出的Hb的多少，从而反映穿孔素溶血活性大小。并以此指标检测肺癌确诊病人。结果 效靶比1∶20，反应体系中加入Ca2+，5% CO2，37℃孵育16小时，可较好反映穿孔素的溶血活性。肺癌患者的PBMC穿孔素溶血活性较正常人明显降低，有肿瘤转移者较无转移者降低。结论 穿孔素溶血活性的测定对肺癌的诊断及癌肿转移的预报具有重要价值。关键词 穿孔素；溶血活性；肺癌
