雌激素受体基因多态性与冠心病及血脂关系的研究

     【摘要】 目的 探讨雌激素受体基因多态性与冠心病和血脂水平是否相关。方法 应用聚合酶链反应(PCR)和限制性片段长度多态性(RFLP)分析方法,检测200例冠心病患者及190例正常对照者ER基因型,并与各组的血脂水平进行比较分析。结果 ER等位基因X、x和P、p频率在冠心病和对照组分别为0.143、0.857, 0.445、0.555和0.140、0.860, 0.424、0.576。基因型频率分布符合hardyweinberg平衡定律。XbaⅠ及PvuⅡ酶切多态性中,基因型频率、等位基因频率及结合XbaⅠ和PvuⅡ两个酶切多态性分析在组内、组间比较差异均无显著意义(P>0.05),且ER基因型在两组间血脂水平比较差异亦无显著意义(P>0.05)。结论 在冠心病患者中存在ER XbaⅠ和PvuⅡ基因多态性,但XbaⅠ和PvuⅡ酶切多态性与冠心病的发生无相关性,与血脂水平亦无相关性。

    A study of the correlation between estrogen receptor gene polymorphism and coronary heart disease as well as serum lipid levels in postmenopausal women

    【Abstract】 Objective To investigate the distribution of estrogen receptor (ER) in Chinese Hans and the correlation between estrogen receptor gene polymorphism and coronary heart disease (CHD) as well as serum lipid levels in postmenopausal women.Methods The genotype in 200 patients with coronary heart disease and 190 healthy women were detected.The PvuⅡ and XbaⅠ restriction fragment length polymorphisms (RFLP) of the ER were anlyzed by polymerize chain reactionrestriction fragment length polymorphism (PCRRFLP).The blood lipid levels in both groups were measured at same time.Results The ER allelic frequencies of X,x and P,p alleles were 0.143,0.857;0.445,0.555 and 0.140,0.860;0.424,0.576 in coronary heart disease group and control group respectively.There was no significant difference in genotype distribution and allele frquencies of PvuⅡ and XbaⅠbetween two groups (P>0.05).There was no significantly difference in total cholesterol (TC),triglyceride (TG),HDLC and LDLC levels among ER genotypes.Conclusion There exsists gene polymorphism of ER XbaⅠand PvuⅡ in patients with CHD,but there is no significant difference in genotype distribution and allele frequencies of PvuⅡ and XbaⅠ between CHD patients and healthy women and there is no significant difference in total cholesterol (TC),triglyceride (TG),HDLC and LDLC levels among ER genotypes. ......
