保肝药物在门静脉高压症犬与正常犬肝血药浓度差异及其临床意义

[摘要] 目的 探讨保肝药物在门静脉高压症犬和正常犬的肝血药浓度的差异，揭示门静脉高压条件下常规保肝治疗效果不佳的部分原因，并提出相应的改进方法。方法 本地健康犬10只采取自身对照的方法，对比门静脉高压模型前后外周静脉及肝动脉给药外周血药浓度及肝血药浓度的差异，计算药-时曲线下面积(areaunder curve,AUC)。结果 对于正常犬经外周静脉给药，其外周血药浓度与肝血药浓度基本一致；经外周静脉(头静脉)给药正常犬的肝血AUC是门静脉高压犬的2.78倍；门静脉高压犬经肝动脉给药其肝血AUC是经头静脉给药的3.76倍。结论 门静脉高压症条件下，通过外周静脉常规剂量治疗不能持续有效地达到肝血药治疗浓度，经肝动脉给药是一条可行的给药途径。[关键词] 门静脉高压；动物模型；犬
Difference of medicine consistency of blood in liver between the portal hypertension and normal canines and clinical value

[Abstract] Objective To explore the difference of protective liver medicine’s consistency in liver between the portal hypertension (PH) and normal canines,and demonstrate the reasons that the curative effect of difference of protective liver medicine isn’t well under the situation of portal hypertension.Methods To contrast the medicine consistency of blood in liver injected by vein of head and hepatic artery before and after PH canines model and calculate the areaunder curve.Results The medicine consistency of blood in liver as same as in vein injected by head vein.It is 2.78 times that the medicine consistency of blood in liver of normal canine injected by vein of head contrast with the PH canine,while it is 3.76 times that the medicine consistency of blood in liver of PH canine injected by hepatic artery contrast with the head vein.Conclusion Under the situation of PHT,to provide routine medicine by peripheral vein can’t make the medicine consistency of blood in liver reach the effective dosage,but the medicine injected by hepatic artery is a hopeful way. ......
