高速离心对藻胆蛋白提取纯化的影响

【摘要】 探索条斑紫菜藻胆蛋白粗提过程中高速离心作用对提纯的影响,为进一步纯化作铺垫。首先分别以高速离心法与超滤法处理藻胆蛋白盐析液,发现在对提纯贡献相近的情况下,前者受溶液量和浓度的限制更小。其次在多步盐析过程中多次插入高速离心操作,结果显示最后一次高速离心即可起到相同的提纯效果,且认为离心与盐析无法相互替代。最后研究了高速离心对脱盐液保存的影响,认为其在沉降大分子物质和变性蛋白的同时,也必然会使目标蛋白有所损失。结果显示适当的高速离心作用在藻胆蛋白粗提阶段可收到较好的效果。【关键词】 藻胆蛋白;离心;脱盐;产率;纯度

【Abstract】 The effects of high-speed centrifugation on extraction and purification of crude phycobiliprotein were studied. High-speed centrifugation was operated firstly to purify protein, which has advantage on little limitation of quantity and concentration of solution compared to filtration. Then it was used during,several ammonium sulphate precipitation showing,that the last high-speed centrifugation was enough and unsubstitutable.
