小鼠骨髓来源树突状细胞的体外扩增及鉴定

【摘要】 目的 体外诱导扩增小鼠骨髓来源树突状细胞(DC),并从形态、表面分子和功能三方面进行鉴定。方法 无菌取骨髓细胞培养4h后,除去悬浮细胞,然后加入rmGM-CSF和rmIL-4培养,7天后观察DC的形态特征,流式细胞仪(FACS),检测DC表面分子及混合淋巴反应(MLR),检测DC刺激同种异体T细胞增殖的能力。结果 培养出的DC具有大小不一的突起,表达小鼠DC特异的表面标志33D1,及表面分子CD40、CD80和CD86;具有强烈刺激同种异体T细胞增殖的能力,且随DC数目的增多而增强。结论 从小鼠骨髓细胞中扩增出大量具有典型特征的DC,为后续研究奠定基础。【关键词】 树突状细胞;骨髓细胞;混合淋巴反应
Amplification and identification of dendritic cells derived from mouse bone marrow in vitro

【Abstract】 Objective To induce and amply dendritic cells (DCs) from mouse bone marrow in vitro, and identify from their morphology,surface molecules and function. Methods Isolated mouse bone marrow cells were cultured 4 hours, then removed suspended cells and added rmGM-CSF and rmIL-4.After 7 days, DCs morphological characterization was observed, the expression of the surface molecules was examined by fluorescent activated cell sorter(FACS), DCs’ power to stimulate the proliferation reaction of allogeneic T cells was detected with mixed lymphocytes reactivation(MLR).Results Cultured DCs had longer or shorter dendrites ;expressed mouse special marker 33D1 and surface molecules including CD40,CD80,CD86;and intensely stimulated the proliferation of allogeneic T cells, What’s more, the power of proliferation was enhanced with the increase of numbers of DCs.Conclusion Obtain a lot of DCs had typical characterization from mouse bone marrow, and establish foundation for further studies. ......
