HPLC法测定奥美拉唑的血药浓度

     【摘要】 目的 建立人血浆中奥美拉唑浓度的HPLC测定方法。方法 以C18柱(250mm×4.6mm,5um,Discovery)为色谱柱,乙腈:0.01mol/L K2HPO4(pH7.5)=35:65(v/v)为流动相,检测波长302nm。结果 奥美拉唑峰形良好,血浆中内源性物质不干扰样品的测定,批内、批间精密度符合要求,在0.02~2μg/ml浓度范围内线性关系良好。结论 本法灵敏度高,专属性强,血样处理方法简便。适于药代动力学研究的要求。

    Determination of omeprazole concentration in plasma by HPLC

    【Abstract】 Objective The aim of the present study was to establish a method which can determine the blood concentration of omeprazole. Methods The RP-HPLC method was used. The analytical column was Discovey C18 (4.6mm×250mm,5μm) and the mobile phase consisted of acetonitrile-0.01mol/LK2HPO4 (35:65,v/v).The pH was adjusted to 7.5±0.1 with phosphoric acid and triethylamine at fiow rate of 1.0ml/min. The detection wavelength for omeprazole was 302nm. Results The substance of plasma did not hinder sample from determining. The linear range was 0.02-2μg/ml.The minimum detection limit was 0.02μg/ml. The inter-day and intra-day RSD was less than 15%. Conclusion Be confirmed, the indexes of the method can up to all the requirement to determine creature samples and it is simple and sensitive. So, it is applicable to determine the concentrations of omeprazole in human plasma. ......
