【Abstract】 AIM: To observe the effect of metabolic correlation factors on ionic transport and cell apoptosis of proximal renal tubular epithelial cells at the cellular level through observing the effects of uric acid and insulin of different concentrations on cAMP and Na+K+ATPase in proximal renal tubular epithelial cell line LLCPK1. METHODS: Proximal renal tubular epithelial cells (LLCPK1) from pig were cultured for 24 h in the medium supplemented with uric acid at 0, 0.1, 0.2, 0.4 mmol/L and insulin at 0, 10-9, 10-8, 10-7 mol/L; and then were observed in the level of cAMP into LLCPK1 cells, the activity of Na+K+ATPase and the results of ionic transport. RESULTS: After LLCPK1 cells were exposed to different concentrations of uric acid and insulin for 24 h,in uric acid group the activity of cAMP,[Ca2+],[Na+]were significantly increased in a concentrationdependent manner, but Na+K+ATPase activity, [K+] dropped in highconcentration insulin groups, [Ca2+] and [Na+] were significantly elevated,but cAMP, Na+K+ATPase and [K+]were decreased. CONCLUSION: Uric acid and insulin could cause the increase of the concentration of calcium ion in cell, which might lead to cell dropsy and even induce apoptosis. In addition, high concentration of uric acid and insulin could do damages to LLCPK1 coordinately. ......
