     Distribution of 99mTcTRODAT1 in brain of rats with Parkinsons disease

    【Abstract】 AIM: To develop 99mTc labeled dopamine transporter (DAT) imaging agent 99mTc(2β [N, N, bis(2mercaptoethyl)ethylenediamino]methyl, 3β(4chlorophenyl)tropane (TRODAT1) for evaluating the changes of DAT in rat models with Parkinsons Disease(PD). METHODS: A total of 40 SD rats were randomly divided into 3 groups: control group (n=14), unilateral PD models (n=14) and multiple cerebral infarction models by injecting autothrombus into carotid artery (n=12). 99mTcTRODAT1 distribution in normal rat striatum was observed. The uptake rates of 99mTcTRODAT1 in healthy side and injured side of PD rat striatum and in two sides of multiple infarction rat striatum were compared. RESULTS: 99mTcTRODAT1 distribution in normal rat brain and striatum exhibited an obvious uptake of 99mTcTRODAT1 in striatum. In PD rats, the uptake was less in injured striatum than that in normal striatum obviously. The uptake rate of multiple infarction rat was similar with that of normal rats. CONCLUSION: 99mTcTRODAT1 might be uptaken specificly by rat striatum, so its imaging can provide a beneficial evidence for the early diagnosis of PD. ......
