高效液相色谱荧光法测定人体内血浆中氟伐他汀的浓度

     [摘要] 目的 建立一种简便、快速的高效液相色谱(HPLC)荧光检测法测定人体内血浆中氟伐他汀的浓度。方法 采用Kromasil C18柱(4.6 mm×150 mm,5 μm),柱温40 ℃,流动相为13 mmol/L溴化四丁基铵-乙腈(51∶49),流速为1.0 ml/min,荧光检测激发波长305 nm,发射波长405 nm。结果 氟伐他汀在1~1000 μg/L范围内线性关系良好(r=0.9994)。日内和日间RSD均低于10%。结论 该方法灵敏度高、快速、简便、无杂质干扰,适合于氟伐他汀的体内分析和人体药动学研究。

    Determination of fluvastatin sodium concentration in human plasma by high performance liquid chromatography with fluorescence detection

    [Abstract] Objective To establish a simple and rapid method for determination of fluvastatin in human plasma by HPLC.with fluorescence detection.Methods Chromatographic column was KromasilC18(4.6 mm×150 mm,5 μm)and 40 ℃ of column temperature.The mobile phase consisted of 13 mmol/L Tetrabutylammonium chloride-acetonitrile(51∶49)with a flow rate of 1.0 ml/min.A fluorescence detector was set at λcx305,λcm405nm.Results The lower limit of qualification was 1 μg/L.The calibration curve was in good linearity over the range of 1~1000 μg/L(r=0.9994).The intar-day and inter-day RSD were less than 10%.Conclusion The method is sensitive,rapid and suitable for the study of pharmacokinetics of fluvastatin. ......
