Significance of Tª²bet/GATAª²3 ratio in immune disbalance of asthmatic rats

    ¡¾Abstract¡¿ AIM: To investigate the significance of the Tª²bet/GATAª²3 ratio in the immune disbalance of the asthmatic rats. METHODS: Twentyª²four male SD rats were randomly divided into a control group and an asthma group (n=12 per group). The expressions of Tª²bet and GATAª²3 mRNA and protein in the lungs were detected by reverse transcriptaseª²polymerase chain reaction(RTª²PCR) and Western blot respectively. RESULTS: In the control group, the expression of Tª²bet mRNA was 0.71¡À0.19, which was significantly elevated compared with that of the asthma group(0.08¡À0.12, P<0.01); the levels of Tª²bet protein expression were 0.72¡À0.22 in the control group and 0.18¡À0.06 in the asthma group (P<0.01); The levels of GATAª²3 mRNA expression were 1.06¡À0.25 in the control group and 1.56¡À0.28 in the asthma group (P<0.01); in the asthma group, GATAª²3 protein expression (1.04¡À0.44) increased compared with that of the control group(2.25¡À0.74, P<0.01); in the control group, the ratio of gene expression of Tª²bet/GATAª²3(0.73¡À0.32) was significantly increased compared with that of the asthma group(0.06¡À0.09, P<0.01), and there was a significant difference between the control group(0.75¡À0.25) and the asthma group(0.09¡À0.04) in the ratio of protein expression of Tª²bet/GATAª²3(P<0.01). CONCLUSION: The mRNA and protein ratios of Tª²bet/GATAª²3 decrease significantly in asthma, which may be one of the objective indexes to evaluate the immune disbalance in asthma. ......
