【摘要】 目的 观察丁酸钠对结肠癌HT29细胞形态和生物学特性的影响。方法 多功能透射显微镜(布氏镜)动态观察丁酸钠作用前后HT29细胞的生长状态、细胞运动及贴壁过程的变化。结果 丁酸钠5 mmol/L作用24 h后,HT29细胞数量明显减少,细胞巢变小,巢内细胞数减少;细胞变成圆形 ,细胞大小趋于一致;细胞核内染色质呈块状聚集,折光性降低,呈现出凋亡细胞形态;贴壁过程明显变缓。结论 丁酸钠可以改变HT29的生长状态,抑制其生长;布氏镜可以动态地观察药物作用前后培养细胞的形态、微细结构及运动的变化,是一项先进的、具有实用价值的检查技术。【关键词】 多功能透射显微镜(布氏镜);丁酸钠;HT29细胞株

【Abstract】 Objective To investigate the effect of sodium butyrate on the appearance and bionomics of colon cancer line HT29.Methods Viewing the effect of sodium butyrate on differences of the growth,as well as the cell movement,and the adherence procedure of HT29 with bradford variable projective microscopy (BVPM).Results After 24 hours of coincubation of HT29 and sodium butyrate (5mmol/L),it was showed that the amount of HT29 decreased,the cell nests shrank,and the cell population of each cell nest diminished;most of the HT29 changed into globular,and the bulk of HT29 looked welldistibuted;chromatin gathered uneven,and the refraction weaken,many of them looked as apoptosis cells;adherence procedure of HT29 stepped down obviously.Conclusion Sodium butyrate may inhibit HT29 growth and monitoring the changes of cultured cells in cytomorph ......
