【Abstract】 AIM: To establish the model of Plasmodium berghei (P. berghei) transfected with Plasmodium falciparum (P. falciparum) apical membrane antigen1 (AMA1) gene. METHODS: The transfection plasmid pDB.DTm.DB./AB.AF.DB. containing P.falciparum AMA1 was enzymed by BglI. Blood stages of P.berghei were cultured in vitro, and mature schizonts were purified. Parasites transfected by electroporation were injected into tail veins of mice. Transfected parasites were selected by treating mice with pyrimethamine. DNA of transfected, resistant parasites was isolated and analyzed by PCR.RESULTS: The gene encoding P.falciparum AMA1 was amplified by PCR in the transfected parasites. CONCLUSION: The model of P.berghei transfected with P.falciparum AMA1 gene was established, which provided a basis for further study. ......
