弥漫性大B细胞淋巴瘤克隆性基因重排检测研究

     [摘要] 目的对弥漫性大B细胞淋巴瘤(DLBCL)进行IgH基因重排研究。方法用免疫组化法标记筛选DLBCL，用针对IgH 单轮扩增引物对DLBCL石蜡包埋组织进行多聚酶链反应(PCR)扩增检测克隆性基因重排。结果 60例DLBCL IgH基因重排阳性率为517%(31/60),其中中心母细胞型阳性率545%(30/55),免疫母细胞型阳性率00%(0/3),间变性大细胞型阳性率500%(1/2)。结论 DLBCL IgH基因重排检出率低于B细胞淋巴瘤总检出率, 可能与DLBCL 中存在IgH可变区体细胞超突变及背景细胞的数量有关。

    [Abstract] Objective To investigate the features of gene rearrangement of immunoglobulin heavy chain (IgH) in diffuse large Bcell lymphoma(DLBCL) Methods 60 cases of DLBCL were selected by immunohistochemical method and IgH clonal gene rearrangement were analysed by PCR from paraffinembeded sections derived from the cases above Results IgH gene rearrangements were detected in 517%(31/60) of DLBCL The positive rate of IgH gene rearrangement in centroblastic, immunoblastic, anaplastic large cell lymphoma was 545%,00% and 500% respectivelyConclusion The positive rate of IgH gene rearrangement in DLBCL is lower than that in total Bcell lymphoma,it may be correlated with somatic hypermutation in IgH variable region and the amount of background cells ......
