氯沙坦对高血压大鼠主动脉平滑肌HBEGF表达的影响

     【摘要】目的探讨肝素结合表皮生长因子样生长因子(HBEGF)在自发性高血压大鼠(SHR)主动脉平滑肌的表达情况及氯沙坦对其影响。方法取16只8周龄SHR，随机分为2组，每组8只。氯沙坦组，给予氯沙坦30 mg/(kg·d)溶于饮水灌胃治疗；SHR阳性对照组给予正常饮水。另有8只同龄同源雄性正常血压Wistar kyoto大鼠(WKY组)作为正常对照组。实验周期12周。逆转录多聚酶链反应(RTPCR)和免疫组化实验检测氯沙坦干预12周后各组大鼠HBEGF的表达情况。结果正常对照组和氯沙坦组血压均低于SHR阳性对照组；SHR阳性对照组HBEGF表达均高于正常对照组和氯沙坦组；氯沙坦组HBEGF表达高于正常对照组。结论肝素结合表皮生长因子在SHR表达明显增加而氯沙坦能部分阻断HBEGF的表达。提示HBEGF可能在高血压发生发展过程中起一定作用。

    Effects of losartan on HBEGF expression in the aortic smooth muscle of SHR

    【Abstract】 Objective To observe the expression of HBEGF in the aortic smooth muscle of spontaneously hypertensive rats (SHR) and the effects of losartan. Methods Sixteen 8weekold male SHR were randomly divided into 2 groups: SHR positive control group and losartan group [30 mg/(kg·d)];Eight 8weekold male Wistar kyoto(WKY) rats served as normal control group. Losartan group was given a daily dose of 30 mg/kg losartan for 12 weeks by gavage and the other two groups were fed with a normal diet.The expression of HBEGF in aorta smooth musle was determined by RTPCR and immunohistochemistry. Resukts Blood pressure increased in the SHR positive control group more than that in the losartan group and normal control group. The expression of HBEGF in SHR control group increased more than that in the losartan group and WKY group. The indexes in the losartan treating group increased more than those in the WKY group. Conckusions Hypertension induces HBEGF expression and losartan can decrease the expression of HBEGF, which is possibly involved in the development of hypertension. ......
