银杏内酯B对内皮细胞损伤的保护作用及机制

     【摘要】 目的 探讨银杏内酯B(GB)对TNFα刺激内皮细胞引起的氧化应激性损伤的保护作用及机制。方法 用分子探针2,7-DCF测定细胞内ROS的产生量以及用Western blot测定ERK2激酶和转录因子AP-1的蛋白表达。结果 TNFα(5ng/ml)作用24h后,脑血管内皮细胞(BVEC)中ROS的产生量明显增加(P<0.05),细胞内AP-1、ERK2的蛋白表达显著增强;但GB(50mg/L)和NAC(20mM)能显著性抑制TNFα诱导的细胞内ROS的产生和AP-1、ERK2的表达(P<0.05)。结论 GB能抑制TNFα诱导的BVEC内ROS的水平,此作用可能是通过抑制ROS介导的ERK2激酶及转录因子AP-1的表达而实现。

    Protective Effect of Ginkgolide B on Endothelial Cells Dysfunction and Mechanisms Concerned

    【Abstract】 Objective To investigate the protective effect of Ginkgolide B(GB)on the endothelial cells against oxidative stress exposed to TNFα and to explore the related mechanisms.Methods The ROS production in the cells was detected by using 2,7-dichlorofluorescin diacetate as probe and the protein expression of the AP-1 and ERK2 determined by Western blot.Results Stimulation for 24h by TNF-α(5ng/ml)caused a remarkably increase in the ROS production in the BVEC(P<0.05),and the protein expression of AP-1 and ERK2 were increased; whereas both GB(50mg/L) and NAC(20mM) significantly inhibited the ROS level and the expressions of AP-1 and ERK2 enhanced by TNFα(P<0.05).Conclusions GB is able to inhibit the ROS production in the TNFα-stimulated brain vascular endothelial cells(BVEC),mainly through the inhibition of the ROS-mediated signaling mechanisms and the expressions of ERK2 and AP-1. ......
