【Abstract】 AIM: To study the effect on CaSki cells by inhibiting human papillomavirus (HPV)16 E6 gene expression in CaSki cell. METHODS: The vectors expressing siRNA of HPV16 E6 gene were constructed and transfected into CaSki cells. The expression of E6 mRNA was detected by RTPCR and the expression of P53 protein by Westen blot. CaSki cell cycle was analyzed by flow cytometry. RESULTS: After transfection, the expression of HPV16 E6 mRNA was reduced and the expression of P53 was upregulated in CaSki cell line. The proliferation of CaSki cells were inhibited and the cells were arrested at G0G1 stage. CONCLUSION: RNAi can inhibite the expression of HPV16 E6 gene in CaSki cells, which upregulates the expression of tumor suppression protein P53 and results in growth retardation of CaSki cells. ......
