comConstruction of DDR2/EGFP fusion vector and analysis of its activity in 293 T cells

    ¡¾Abstract¡¿ AIM: To find a way of detecting the ectopic expression of discoidin domain receptor 2(DDR2) in the cell and of tracing dynamically the expression distribution of DDR2 protein. METHODS: The fusion construct of DDR2 with EGFP was obtained by PCR amplication and restriction enzyme digestion, and transfected into 293T cells. The expression of the green fluorescence protein was examined by inverted microscope and the expression and activation of this fusion protein was confirmed by Western blot. RESULTS: DDR2/EGFP fusion plasmid was successfully constructed. Further analysis also demonstrated that the fusion protein has similar expression and activation pattern with wild type ones. CONCLUSION: The success in constructing the fusion plasmid DDR2/EGFP and in detecting its phosphorylation can lay a foundation of studying the function of DDR2. ......
