Expression of runx3 mRNA and protein in gastric carcinoma and its correlation with clinicopathological parameters

    ¡¾Abstract¡¿ AIM: To investigate the expression of runx3 gene mRNA and protein in human gastric carcinoma and surrounding normal tissue and its correlation with clinicopathological parameters. METHODS: RNA and protein were extracted from 60 human gastric carcinoma and surrounding normal tissue specimens. Reverse transcription polymerase chain reaction (RTª²PCR) and Western blot were used to detect the runx3 mRNA and protein level. RESULTS: The relative expression value of runx3 mRNA found in human gastric carcinoma specimens was 0.7542¡À0.1284, and that did in surrounding normal tissue specimens was 0.8427¡À0.0541(t=4.941, P<0.0001). The integral optical density value of runx3 protein confirmed in human gastric carcinoma specimens was 10828¡À989.9 and that did in surrounding normal tissue specimens was 47833¡À7566 ( t=35.57, P<0.0001). RTª²PCR and Western blot indicated expression of runx3 in gastric carcinoma was significantly lower than that in corresponding normal stomach tissue respectively. CONCLUSION: Lower expression of runx3 gene mRNA and its protein in the gasric carcinoma probably plays an important role in the tumorigenesis and development of gastric carcinoma. ......
