[摘 要] 目的: 建立稳定的大鼠原位肝移植模型，为进一步研究核苷酸圈套技术抗大鼠肝移植急性排斥建立良好 的技术平台。方法: 实验以SD大鼠为模型动物，采用改良“二袖套管法”进行原位肝移植实验，统计受体大鼠术后生存时间，并常规病理检测术后移植肝病理变化。 结果: 共进行大鼠原位肝移植实验82次。实验后期能稳定建立起原 位肝移植模型，供体手术时间25～35 min;供肝冷缺血时间40～55 min;受体无肝期时间13～24 min;受体手术时间45～65 min;2周生存率为93%。移植肝组织在术后1周可见大量的单核淋巴细胞浸润，在门静脉周围的汇管区尤为明显;肝脏实质有点状坏死。术后2周细胞浸润增加，肝实质可见桥状坏死。术后1月移植肝组织内细胞浸润和坏死减少，且可见肝细胞和胆管增殖。 结论: 采用改良“二袖套管法”建立大鼠原位肝移植模型手术成功率高，稳定性好，是研究肝移植术后机体生化生理改变、免疫状态变化等的良好技术平台。
