【Abstract】 AIM: To observe the effects of octylphenol (OP) on cell cycle and cyclin expression of MDAMB231 breast cancer cells and to probe the molecular mechanisms of OP inhibiting MDAMB231 breast cancer cell proliferation. METHODS: The changes of cell proliferation, cell cycle, cyclinD1 mRNA, cyclinD3 mRNA, CDK2 mRNA, CDK4 mRNA and cyclinD1 protein expressions of MDAMB231 cells treated with OP were observed by using MTT test, flow cytometry, immunocytochemistry and RTPCR. RESULTS: When MDAMB231 breast cancer cells were treated with 4 or 8 μmol/L OP for 48 h, the cell inhibition rates were (17.13±4.1)%, (40.25±8.7)%, and the fluorescence values of cyclin D1 were 55.1±10.2, 41.4±11.2, significantly decreased as compared with control group(89.9±11.2, P<0.05). When MDAMB231 breast cancer cells were treated with 8 μmol/L OP for 24 and 72 h, the cell ratios in the G0/G1 phase were (67.0±17.6)% and (44.4±11.9)% ......
