Correlation analysis between GFPT2 gene polymorphisms and type 2 diabetes in Han population in Shanghai of ChinaWANG HongLing, YU YongChun, LI Zhi, WANG YanBo, LI JingHua, LIU Rui

【Abstract】 AIM: To evaluate the distribution of the GFPT2 gene polymorphisms IVS2+26G>A, rs2303008, rs2303007 and rs1035411 and their associations with type 2 diabetes (T2DM) in Han population of Shanghai city of China. METHODS: Four common SNPs of GFPT2 gene were genotyped with PCRRFLP and then analyzed with transmissiondisequilibrium test (TDT) and sib transmissiondisequilibrium test (STDT) in 75 T2DM pedigrees (totally 335 individuals, among them 196 were T2DM patients), and haplotypes combined by the four SNPs were calculated by Genehunter software. Furthermore, the association of GFPT2 polymorphisms with physiological and biochemical parameters was also studied. RESULTS: (1) There was no positive result in four variants in TDTSTDT analysis (P>0.05). (2) There was no preferential transmission of any haplotype (P>0.05). (3) There was no difference in physiological and biochemical parameters between genotypes and allele genes of four SNPs, respectively (P>0.05). CONCLUSION: The four variants of GFPT2 gene were not associated with T2DM in TDTSTDT analysis, and no significant association of haplotypes with T2DM and IGT was detected in our study. The studied SNPs in GFPT2 gene may not be major susceptible sites of T2DM in Han population in Shanghai. ......
