摘要:目的获取高质量的岩黄连基因组DNA。方法用改进的SDS-CTAB 法从岩黄连叶片中提取基因组DNA,通过琼脂糖凝胶电泳、紫外分光光度法、PCR 检测其质量,并与传统的CTAB 法比较。结果通过改进的SDSCTAB 法提取的DNA优于CTAB 法提取的DNA,电泳条带清晰,纯度高,能较好的进行PCR。结论改进的SDSCTAB 法适合岩黄连基因组DNA 的提取。关键词:岩黄连; 基因组DNA; SDSCTAB 法
Isolation and Analysis of Genomic DNA from Leaves of Corydalis saxicola Bunting

Abstract:ObjectiveIn order to obtain high quality genomic DNA of Corydalis saxicola Bunting,an effective method was established. MethodsTwo different methods,CTAB method and improved SDSCTAB method, were applied to isolate genomic DNA from leaves of C. saxicola. The DNA obtained was analyzed by the means of agarose gel electrophoresis, spectrophotometer measurement and PCR.ResultsHigher purity DNA was obtained by improved SDSCTAB method and it showed good response to PCR. ConclusionThe improved SDSCTAB method was favorable for the extraction of genomic DNA from C. saxicola. ......
