丹参单体对TNF-α诱导大鼠动脉平滑肌细胞MCP-1和IL-1β的影响 (pdf)【摘要】 目的 观察丹酚酸B和丹参酮ⅡA对肿瘤坏死因子(TNF-α)诱导的血管平滑肌细胞(SMC)白细胞介素-1β(IL-1β)和单核细胞趋化蛋白-1(MCP-1)的影响,探讨丹参单体抗动脉粥样硬化(AS)的作用机制。方法 体外培养大鼠主动脉SMC;RT-PCR法检测SMC MCP-1 mRNA的表达;放免法检测SMC培养上清IL-1β的水平。结果 TNF-α促进SMC MCP-1mRNA的表达,提高IL-1β水平(P值均<0.01)。两种丹参单体均抑制上述因子的表达,其中,对于抑制MCP-1mRNA的表达以丹参酮ⅡA效果更佳。结论 丹参单体抗AS作用的机制之一是抑制炎症相关因子的表达,丹参酮ⅡA抗炎效果优于丹酚酸B。
