摘要:笔者回顾了10多年来白屈菜红碱诱导细胞凋亡的细胞和分子机理的研究,总结这些机制包括白屈菜红碱对BclXL(Bcl2家族的抗凋亡成员)和PKC的复杂作用,促进Cytc的释放和ROS的产生以及对微管蛋白的抑制作用;同时简要地介绍了相关的实验。关键词:白屈菜红碱; 细胞凋亡
Review on the Mechanism of Apoptosis Induced by Chelerythrine

Abstract:This article reviews recent ten years' investigations in the molecular and cellular mechanisms of actions by which chelerythrine induces cells apoptosis, summarizes the mechanisms including complex functions of chelerythrine on BclXL (an antiapoptosis member of Bcl2 family) and PKC, promotions CytC release and ROS generation and inhibition on tubulin polymerization. It also briefly introduces the relative tests.

白屈菜红碱属于苯并菲啶类碱,其氯化物分子分子式是 C21H18NO4Cl,分子量是383.84,能溶解于水和DMSO。对它的药理作用的研究由来已久。Steven 等[1~7]在体外实验中证实白屈菜红碱对9种肿瘤细胞(HT29,MCF7,MCF7ADR,DaOY,SQ20B,SCC61,JSQ3,SCC35,LnCaP)表现细胞毒性。细胞通过不同浓度的白屈菜红碱处理在37°C下孵育4 h,其ED50的范围是2~5 μm[8]。对化学和放射耐受的肿瘤细胞株包括SQ20B, JSQ3,SCC35(都是p53突变型)和对放射敏感的SCC61表现了与其它相同的剂量-效应曲线 ......
