摘要:目的探讨高效液相色谱(HPLC)法建立β细辛醚药代动力学的参数设置及生物样品制备条件筛选。方法在石菖薄挥发油β细辛醚在大鼠的药代动力学实例研究的基础上,对给药剂量、血样采集时间点、生物样品的贮存稳定性、蛋白沉淀剂的种类及剂量、样品提取和制备方法,以及流动相、波长、柱温等参数进行了比较。结果蛋白沉淀的种类及剂量、样品提取和制备方法对方法回收率及灵敏度影响较大;流动相、色谱柱、波长与流速等对分离度及保留时间影响较大。柱温以及样品温度影响较小。结论HPLC法建立β细辛醚药代动力学的方法学可靠、准确、回收率高。关键词:β细辛醚; 药代动力学; 方法学; 色谱条件

Abstract:ObjectiveTo discuss the condition of HPLC and sample preparation of βAsarone pharmacokinetics.MethodsBase on pharmacokinetics research of βasarone in volatile oil of Acorus tatarinowii Schott in rat serum,to compare the condition of HPLC and sample preparation.ResultsThe main influence factors included mobile phase,chromatogram column,detection wavelength,way of sample preparation,kind and dose of proteinic precipitator.Temperature of column and sample were unconspicious.ConclusionMethod of HPLC βasarone pharmacokinetis is accurate and with high recovery. ......
