AG490对SD/Waistar大鼠心脏移植物的免疫抑制作用

     摘要: 目的 观察Jak/STAT阻断剂(AG490)作为免疫抑制剂在动物大器官移植模型中的效果,初步探讨其作用机制。方法 建立SD/Waistar大鼠心脏移植模型,移植当日起予AG490和/或环孢素A,共8 d,观察移植物存活时间,测定外周血IL2、IL6水平。结果 AG490能显著延长移植物存活时间,与环孢素A联用有协同效应,作用更强。AG490降低大鼠移植后血清IL2水平。结论 AG490阻断Jak/STAT通路可起到免疫抑制的效果,该机制可能与降低血清IL2水平有关。

    Effects of AG490 on Outcome in SD/Wistar Heart Transplantation

    ABSTRACT: Objective To evaluate the possibility and prospect of the AG490 as a potential immunosuppressor. Methods The rat model of heart transplantation were built in thirty SD rats. All recipients(Wistar) has been separated into four groups, cyclosporin(CsA, 20 mg/kg), AG490(20 mg/kg), CsA+AG490 and sodium chloride(0.2 mL) has been injected into the abdominal cavity separately for 7 days. The graft survival time has been recorded. Results The AG490 prolonged the graft survival time almost equal to CsA, when used both AG490 and CsA the graft survival time has been prolonged more. Othermore, the level of IL2 in rat has been reduced by AG490. Conclusion AG490 can cause immunosuppressive effects and there is synergetic effect when combined with CsA. ......
