     摘要：目的探讨花边莲提取液对人肺癌SPCA1细胞凋亡相关基因survivin和caspase3表达的影响。方法采用体外培养人肺癌SPCA1细胞，实验随机分为正常对照组、花边莲处理组(浓度分别为12.5，25和50 ml/L)和顺铂(DDp25 mg/L)组，半定量RTPCR检测survivin和caspase3 mRNA表达水平，二步法免疫组化检测Survivin和Caspase3蛋白表达变化。结果与正常对照组相比，花边莲各浓度组caspase3 mRNA和蛋白表达显著上升(P<0.01)，survivin mRNA和蛋白表达明显下降(P<0.01)。结论花边莲提取液诱导SPCA1细胞凋亡的分子机理可能与上调caspase3基因表达和下调survivin基因表达有关。

    Abstract：ObjectiveTo investigate the influence of HuaBianLian(HBL) extract on expression of apoptosis associated genes survivin and caspase3 in human lung cancer SPCA1 cells.MethodsCultured human lung cancer SPCA1 cells were randomly divided into the control group (the negative control group),HBL-treated groups(12.5,2.5,50 ml/L) and DDP(25 mg/L) group (the positive control group).The expression of survivin and caspase3 mRNA was detected by semiquantitive RTPCR .The expression of survivin and caspase-3 proteins were detected by twostep immunhistochemical staining.ResultsCompared with control group ,the expression of caspase 3 mRNA and protein increased significanty (P<0.01),while survivin mRNA and protein decreased markedly in HBL groups (P<0.01).ConclusionThe molecular mechanism of HBL apoptosis inducing of SPCA1 cells may be associated with the upregulating expression of caspase-3 gene and downregulating expression of survivin gene. ......
