摘要:目的探讨花边莲提取液对人肺癌SPCA1细胞凋亡相关基因survivin和caspase3表达的影响。方法采用体外培养人肺癌SPCA1细胞,实验随机分为正常对照组、花边莲处理组(浓度分别为12.5,25和50 ml/L)和顺铂(DDp25 mg/L)组,半定量RTPCR检测survivin和caspase3 mRNA表达水平,二步法免疫组化检测Survivin和Caspase3蛋白表达变化。结果与正常对照组相比,花边莲各浓度组caspase3 mRNA和蛋白表达显著上升(P<0.01),survivin mRNA和蛋白表达明显下降(P<0.01)。结论 花边莲提取液诱导SPCA1细胞凋亡的分子机理可能与上调caspase3基因表达和下调survivin基因表达有关。关键词:花边莲; SPCA1细胞; 细胞凋亡; survivin; caspase3

Abstract:ObjectiveTo investigate the influence of HuaBianLian(HBL) extract on expression of apoptosis associated genes survivin and caspase3 in human lung cancer SPCA1 cells.MethodsCultured human lung cancer SPCA1 cells were randomly divided into the control group (the negative control group),HBL-treated groups(12.5,2.5,50 ml/L) and DDP(25 mg/L) group (the positive control group).The expression of survivin and caspase3 mRNA was detected by semiquantitive RTPCR .The expression of survivin and caspase-3 proteins were detected by twostep immunhistochemical staining.ResultsCompared with control group ,the expression of caspase 3 mRNA and protein increased significanty (P<0.01),while survivin mRNA and protein decreased markedly in HBL groups (P<0.01).ConclusionThe molecular mechanism of HBL apoptosis inducing of SPCA1 cells may be associated with the upregulating expression of caspase-3 gene and downregulating expression of survivin gene. ......
