摘要:目的观察圣和散对脑胶质瘤C6细胞株体外生长及诱导分化的作用。方法用圣和散处理脑胶质瘤C6细胞,采用甲基噻唑基四唑(MTT) 比色法检测圣和散对C6 细胞增殖的抑制作用, 以免疫组化SABC法检测胶质纤维酸性蛋白(GFAP)和c-myc表达。结果圣和散可显著抑制 C6 细胞体外增殖,并呈现时间、剂量依赖性。在终浓度为5 mg/ml和10 mg/ml时,对 C6 增殖的抑制率达 (49.62±1.13) %和 (69.38±1.42) %,明显优于对照组(P<0.01)。免疫组化可见 SHP组较对照组 GFAP 表达明显增强,c-myc表达下降。结论圣和散对胶质瘤 C6 细胞系的增殖有明显的抑制作用,能通过诱导分化降低脑胶质瘤细胞恶性程度。关键词:圣和散; 脑胶质瘤; 分化
Inducing Differentiation Effect of Shenghe Powder on C6 Brain Glioma Cells

Abstract:ObjectiveTo investigate the effects of Shenghe Powder(SHP) on the proliferation and inducing differentiation of brain glioma C6 cell line in vitro.MethodsBrain glioma C6 cell line was co-cultured with SHP.Methyl thiagoly tetragolium (MTT) was used to measure the level of the proliferation of C6 cultured with SHP.The expressions of glial fibrillary acidic protein (GFAP) and c-myc of C6 cells were detected by immunohistochemical SABC method. ResultsThe proliferation of C6 was obviously inhibited by SHP in a concentration- and time-dependent manner. The inhibitory rate was (49.62±1.13) % and (69.38±1.42) % in SHP at 5mg/ml and 10 mg/ml,compared with the control group, there was a significant difference(P<0. 01) .The expression of GFAP was obviously enhanced and c-myc decreased significantly in C6 cells cultured with SHP.ConclusionSHP can inhibit the proliferation of C6 and decrease malignant degree by inducing differentiation of the C6 cells . ......
