摘要：目的探讨羊胎素对体外培养的SD大鼠骨髓基质细胞(Marrow Stroma Cells, MSCs)的影响。方法用MTT法检测MSCs的增殖；PNPP偶氮法检测MSCs碱性磷酸酶(ALP)的活性；细胞ALP染色观察成骨细胞的形成。结果在含羊胎素原液的0.625%，1.25%，2.5%和5%时对体外培养的SD大鼠MSCs有明显的促进增殖的作用(P<0.01);在含羊胎素原液的2.5%及5%+成骨诱导剂时， ALP的活性明显高于诱导剂对照组(P<0.05)，而在含羊胎素原液的2.5%和5%浓度组， ALP的活性明显高于空白对照组及诱导剂对照组(P<0.001)；细胞ALP染色呈强阳性，加药组颜色明显深于对照组和诱导剂对照组。结论 羊胎素对体外培养的SD大鼠MSCs的增殖和向成骨细胞分化具有明显的促进作用。关键词：羊胎素； 骨髓基质细胞； 成骨细胞； 增殖； 分化
Effects of Lamb Placentin on Marrow Stroma Cells Proliferation and Osteogenic Differentiation in vitro

Abstract：ObjectiveTo investigate the effects of placentin, the substance extract from lamb placenta, on SD rat's marrow stroma cells ( MSCs) proliferation and osteogenic differentiation in vitro. MethodsProliferation of MSCs mediated by lamb placentin was measured by MTT assay. The alkaline phosphatase (ALP) activity of MSCs was detected by PNPP assay. Meanwhile, the cells osteogenic differentiation was examined by ALP staining.ResultsThe growth of MSCs was obviously accelerated by the lamb placentin at the concentration of 0.625%, 1.25%, 2.5%, 5% (P<0.01); the activity of ALP of MSCs at the groups of 2.5% and 5% placentin containing osteoblast inducer was significantly higher than that of the blank group (P<0.05). However, the activity of ALP at the groups of 2.5% and 5% lamb Placentin without osteoblast inducer was significantly higher than the blank group and osteoblast inducer control group (P<0.001). The ALP staining cells at all placentin groups exhibited strong positive results. The colors of the cells at those groups were deeper than the inducer control group. ConclusionThe lamb placentin can remarkably increase the proliferation and induce the osteogenic differentiation of MSCs. ......
