摘要:目的研究黄芩茎叶总黄酮(SSTF)对乙醇损伤小鼠原代培养肝细胞的保护作用。方法采用胰蛋白酶消化、分离小鼠肝细胞进行原代培养,乙醇体外诱导肝细胞损伤,以不同浓度的SSTF对肝细胞保护,检测培养液中丙氨酸氨基转移酶(ALT)活性和肝细胞内丙二醛(MDA)含量,MTT法测肝细胞存活率。结果SSTF(100,200,300 mg·L-1)明显改善肝细胞存活率,抑制乙醇引起的ALT活性的升高,对肝细胞中MDA含量的升高有明显的抑制作用。结论SSTF对乙醇损伤的小鼠肝细胞有明显的保护作用,其机制可能与SSTF的抗氧化作用有关。关键词:乙醇; 肝细胞; 原代培养; 黄芩茎叶总黄酮; 保护作用
The Protective Effect of Total Flavonoid in scutellaria baicalensis Stem-Leaf on Primary Cultured Mice Hepatocytes Injured by Ethanol

Abstract:ObjectiveTo study the protective effect of total flavonoid in Scutellaria baicalensis stem-leaf (SSTF) on primary mice hepatocyes injured by ethanol. MethodsPrimary hepatocytes were isolated, cultured by trypsin digestion and induced by ethanol. Various concentrations of SSTF were added to the primary cultured hepatocytes for 12h.The content of malondialdehyde(MDA) in hepatocytes and the activity of ALT in cultural supernatant were determined by general methods,the viabilities of hepatocytes were measured by MTT.ResultsThe elevation of MDA content of hepatocytes and ALT level in supermatant of cultural hepatocyes were restored remarkably by SSTF, hepatocytes viability was improved significantly by SSTF.ConclusionSSTF has a protective action on primary mice hepatocyes injured by ethanol.This might be associated with its anti-lopoperoridation. ......
