砷暴露小鼠肝组织核酸损伤免疫组织化学观察

摘要:目的 为进一步研究砷对肝脏的毒性作用机制提供实验依据。方法 昆明种小鼠40只,随机分为4组:1,2,4mg/L三氧化二砷染毒组和生理盐水组,连续染毒60d,取小鼠肝组织固定,用免疫组化方法观察肝脏组织细胞的8Nitroguanine表达。结果 染砷组小鼠肝细胞出现抗8Nitroguanine免疫染色阳性反应,并随着砷暴露剂量的增加 而呈现明显的剂量-反应关系。结论 慢性低剂量砷暴露可诱发小鼠肝细胞8Nitroguanine的高表达,提示8硝基鸟嘌呤核苷(8Nitroguanine)可以作为砷对肝细胞毒性作用的早期敏感生物标志物。关键词:砷;肝;8Nitroguanine;免疫组化;核酸损伤
Observation on nucleic acid damage of liver in mice exposed to arsenic in low dose by immunohistochemistry

Department of Hygiene,Dalian Medical University(Dalian 116027,China)Abstract: Objective To provide evidence for exploring the mechanism of arsenicinduced hepatoxicity.Methods Forty mice were diveded into 4 groups, e.g.,one controls and 3 experimental groups(1 mg/L As2O3,2 mg/L As2O3 and 4 mg/L AS2O3).As2O3 was administered for 60 days.Immunohistochemistry method was used to investigate 8Nitroguanine expression in liver tissue of mice.Results Expression of 8Nitroguanine was observed in the liver tissue of mice exposed to arsenic and there was relationship of doseresponse in expression of 8Nitroguanine with the increase arsenic dose.Conclusion Arsenic induced high expression of 8Nitroguanine in liver tissue of mice.8Nitroguanine might be an earlier sensitive bio-marker for arsenic toxicity to liver. ......
