摘要：目的为获得高质量的何首乌总基因组DNA。方法采用CTAB法提取，对其提取方法进行改进。结果用改进的方法提取的样品DNA的OD260/OD280值在1.630~1.955之间，琼脂糖凝胶上主带清晰、降解较少，能完全被限制性内切酶EcoR Ⅰ和MseⅠ完全酶切，得到合适的AFLP指纹样式。结论 改良的CTAB法提示提取何首乌DNA的理想方法，适合AFLP-PCR反应和其他分子生物学应用。关键词：何首乌； DNA提取； AFLP
Genomic DNA Extraction from the Polygonum multiflorum Thunb.

Abstract：ObjectiveTo isolate high-quality genomic DNA from Polygonum multiflorum Thunb.MethodsCTAB isolation method was used and modified in this paper.ResultsUsing the modified CTAB methods, the value of OD260/OD280 of DNAs were between 1.630 and 1.955. Strips were clear and less decomposed in agarose electrophoresis. The DNAs were completedly digested with restriction endonucleases and amplified by the polymerase chain reaction(PCR). Sharp fingerprint patterns of AFLP were obtained.ConclusionThe improved CTAB method was optimal for analysis of AFLP-PCR and other molecular biology research. ......
