摘要: 目的 研究创伤性脑损伤(TBI)后脑组织cfos表达及神经细胞凋亡,探讨神经细胞凋亡在脑继发性损害中的作用,及cfos基因表达与凋亡的关系。方法 雄性Wistar大鼠,随机分为假手术对照组和脑创伤后3、12、24、72小时和7天组,采用自由落体撞击致重型颅脑损伤。应用TUNEL法检测神经细胞凋亡,免疫组化法观测cfos的表达。结果 TBI后3小时创伤组各组脑组织即出现cfos表达和神经细胞凋亡,随伤后时间逐渐增强,至伤后72小时达高峰,伤后7天仍有明显的cfos表达和神经细胞凋亡。其分布区域为脑挫伤区、挫伤周围皮质和海马,累及细胞类型包括神经元和胶质细胞。cfos表达与神经细胞凋亡在时序空间分布上基本一致,两者呈正相关。结论 创伤性脑损伤可引起脑组织明显的cfos表达和神经细胞凋亡,cfos表达增加在神经细胞凋亡和修复中可能起重要作用。关键词:脑损伤;基因;细胞凋亡;神经细胞
Cfos expression and apoptosis after traumatic brain injury

Abstract: Objective To study cfos expression and apoptosis after traumatic brain injury (TBI) in rats,and to explore the role of apoptosis in the secondary brain injury,and the relation between cfos expression and neuroglial apoptosis.Methods Male Wistar rats were randomly divided into the control group and 3,12,24,72h and 7 day group after injury.Parietal brain contusion was made by weightdropping method.Apoptosis was detected by immunohistochemical staining(TUNEL),and cfos expression was detected by immunohistochemistry.Results cfos expression and apoptosis was found in the injury group 3h after TBI,which increased gradually at 12,24,72h and 7d after TBI.The involved cells included neurons and glial cells.cfos expression and neuroglial apoptosis appeared to be similar in temporal changes and spatial localization,with a high relation between these 2 variables.Conclusion It is highly suggested that TBI could induce significant cfos expression and apoptosis after TBI,and cfos expression might be highly responsible for the apoptosis,proliferation and renovation after TBI. ......
