【摘要】 目的 探讨增殖细胞核抗原(PCNA)反义寡核苷酸对大鼠胶质瘤生长的抑制作用。 方法 所有大鼠均在立体定向导引下右尾状核区接种1.0×106胶质瘤细胞。治疗组接种C6胶质瘤细胞后24 h内用PCNA反义寡核苷酸原位穿刺注射进行治疗,对照组只注射Hank’s液,每周1次,连续4周;观察两组大鼠肿瘤的一般情况与生存期。4周后断颈处死大鼠,完整地取下肿瘤,测量肿瘤体积,计算其抑瘤率。 结果 PCNA-ASODN治疗组大鼠胶质瘤的生长明显受到抑制,抑瘤率为64.5%。 结论 PCNA反义寡核苷酸对胶质瘤的生长具有抑制作用,通过反义技术可抑制靶基因的表达,从而抑制胶质瘤细胞的增殖。【关键词】 PCNA;反义寡核苷酸;胶质瘤
The experimental study of PCNA antisense oligonucleotides on inhibiting rat glioma growth

【Abstract】 Objective To study the inhibitory effect of PCNA antisense oligonucleotides on the growth of rat glioma. Methods 1.0×106 glioma cells were cultured and seeded into right caudate nucleus of all rats with stereotactic technique. The treated group was given PCNA-ASODN once a week at 24 hours after cells inoculation . The control group was just treated with Hank’s liquid once a week for four weeks. The volume of tumors were measured. The inhibition rat of tumor growth was calculated. Results The tumor growth of PCNA-ASODN group(2.68±0.48 mm3) was significantly inhibited.The inhibition rate of tumor growth of the PCNA-ASODN group was 64.5%. Conclusion It was demonstrated that the growth of glioma in rat can be inhibited by PCNA antisense oligonucleotides, the expression of PCNA target gene and prolification of glioma cells can be inhibited by PCNA antisense oligonucleotides. ......
