钆喷酸葡胺脂质体的制备与研究

【摘要】 本文采用改良逆相蒸发法-膜挤压法相结合将水溶性顺磁性造影剂钆喷酸葡胺制成小单室脂质体(SUV),方法简单、重现性好,显示了可进一步工业化的良好前景。激光散射法测得脂质平均粒径为195nm。4℃避光放置6个月,各项理化指标基本无变化。对制剂的急性毒性研究结果表明,钆喷酸葡胺及钆喷酸葡胺脂质体的ivLD50值分别为8.00mmol/kg和9.56mmol/kg,这提示脂质体可降低药物的毒性。而影像学结果进一步证明了钆喷酸葡胺脂质体具有靶向性。【关键词】 钆喷酸葡胺;脂质体;制备研究
Research and preparation of liposomal Gd-DTPA

【Abstract】 We used a modified reverse-phase rotary evaporate-extrution method to obtain liposomes containing Gd-DTPA. We had developed a modified one of this technique. The method was simple and good reproduce. It was ideally suitable for scaling-up for production purpose. The mean diameter of liposomes was 195nm, which was determined by laser scattering,this liposomal formulation had no evident effects under 4℃. The iv LD50 of free and liposomal Gd-DTPA was 8.00mmol/kg and 9.56 mmol/kg, respectively. This suggested that Gd-DTPA liposomes had a proven effect in reducing toxicity. Result of imaging confirmed liposomal Gd-DTPA possesed target for liver and spleen. ......
