【Abstract】 AIM: To investigate the effect of antisense oligonucleotides (ASODN) encoding telomerase reverse transcriptase (TERT) mRNA on thyroid carcinoma (TC) cell apoptosis. METHODS: The ASODN and sense oligonucleotides (SODN) encoding TERT were delivered respectively to the TC cells in vitro. The effect of ASODN encoding TERT mRNA on TC cell apoptosis was detected by transmission electron microscope, Hoechest 33258 staining and flow cytometry. RESULTS: Transmission electron microscope showed ASODN could induce typical apoptotic manifestations of TC cells; Hoechest 33258 staining showed ASODN could increase the number of apoptotic cells from 52±16 to 178±32; flow cytometry revealed that the apoptosis rates of TC cells treated by 5 μmol/L ASODN, SODN, or nothing were 10.4%, 1.6% and 0, respectively (P<0.05). CONCLUSION: ASODN complementary to TERT mRNA at some concentration can sequencespecifically enhance TC cell apoptosis. ......
