     [摘要] 纯化是研究病毒理化特性、生物学和免疫学特性的重要步骤和要求。在纯化病毒的方法中,氯化铯(CsCl)密度梯度离心法是较为常用的方法之一,方法简便易行,获得的病毒样品较纯。本文报道一种轮状病毒(RV)的CsCl密度梯度离心纯化方法,在离心前用聚乙二醇(PEG 6000)浓缩得到的病毒增殖液中加入56%CsCl,使其成为均匀混合液,经水平转头密度梯度离心后,可以将病毒分成3条乳白色区带。在密度梯度离心前,病毒样品是否用三氟三氯乙烷抽提,结果也有差别。经SDS-PAGE、Western、病毒的感染性滴度测定结果表明,所纯化的病毒具有较高的纯度和很好的回收率。

    Purification of rotavirus by CsCl gradient centrifugation

    [Abstract] Objective Purification is an important procedure in study on virus physical and chemical properties.Among them,CsCl density gradient centrifugation is often used,because product purity is good by this method.This paper described a method to purify rotavirus from cultural medium by means of CsCl gradient centrifugation.Methods The purified virus samples were further determined by means of SDS-PAGE,Western Blot,and antigenicity detection.Results The results showed that this is a good method with a good purity and recovery rate,and purification were affected 1.1.2-Trichloro-Trifluoroethane extraction before centrifugation.Condusion Good purity and recover’y rate could be obtained by the method. ......
