汉防己甲素、川芎嗪和苦杏仁甙对人肾成纤维细胞的影响

中华肾脏病杂志000312摘 要:目的观察汉防己甲素、川芎嗪和苦杏仁甙3种中药成分对人肾成纤维细胞(KFB)的影响。方法采用ELISA法、四甲基偶氮唑蓝(MTT)法、流式细胞仪、免疫组织化学法分别检测汉防己甲素、川芎嗪、苦杏仁甙对人KFB分泌的Ⅰ型胶原酶活性、人KFB增殖、凋亡、Ⅰ型胶原表达的影响。结果汉防己甲素、川芎嗪、苦杏仁甙3种中药提取成分在各自的最佳浓度范围和作用时间内均能提高人胎KFB分泌的Ⅰ型胶原酶活性、抑制人胎KFB增殖和Ⅰ型胶原的表达、促进人胎KFB凋亡。结论汉防己甲素、川芎嗪、苦杏仁甙在预防及逆转肾间质纤维化中起重要作用。

Abstract:Objective To study the effects of tetrandrine, tetramethylpyrazine and amygdalin on activity of collagenase secreted by human kidney fibroblasts(KFB), proliferation, apoptosis and expression of type Ⅰ collagen of human KFB. Methods Activity of type Ⅰ collagenase secreted by KFB, apoptosis, proliferation and collagen Ⅰ expression of KFB were detected by ELISA, flow cytometer, MTT and immunohistochemical method respectively. Results Tetrandrine, tetramethylpyrazine and amygdalin in their respectively optimal concentration range and effective time range enhanced the activity of collagenasc secrcated by KFB, inhibited proliferation and expression of type Ⅰ collagen of KFB and promoted KFB apoptosis . Conclusion Tetrandrine, tetramethylpyrazine and amygdalin may play important roles in prevention and reversion of renal interstitial fibrosis.
