滴露消毒药水杀菌性能的实验观察

滴露消毒药水杀菌性能的实验观察 提要 滴露消毒药水为含对氯间二甲苯酚(4.8%)、表面活性剂、香料等成份的复方消毒剂。在悬液中，以其1∶400稀释液对大肠杆菌、金黄色葡萄球菌作用10min，杀灭率达99.9%。以其1∶100稀释液浸泡手2min，可使皮肤表面自然菌减少90%以上。10%小牛血清对其杀菌效果略有影响。该剂于37℃贮放3个月，杀菌效果无明显变化。

Abstract
Dettol is a compound disinfectant containing parachlorometaxylenol (4.8%), surfactant, perfume, etc. Suspension test indicated that the killing rate of both Escherichia coli exposed to its 1∶400 dilute solution for 5 min and Staphylococcus aureus exposed to the same solution for 10 min was 99.9%. Immersion of hands in its 1∶100 dilute solution for 2 min could reduce the natural bacteria on skin surface by more than 90%. 10% calf serum had slight influence on its bactericidal efficacy. Its bactericidal efficacy did not change obviously after storing at 37℃ for 3 months.

滴露(Dettol)消毒药水(下简称滴露)主要成分为对氯间二甲苯酚、表面活性剂、香料等。原液含4.8%对氯间二甲苯酚。现将对其杀菌效果与稳定性的实验室观察结果报道如下。
1方法
1.1菌悬液的制备
试验菌为大肠杆菌(8099)、金黄色葡萄球菌(ATCC6538)。取其第3～14代普通营养琼脂24h新鲜培养物，用0.03mol/L磷酸盐缓冲液(pH7.2)稀释成含菌量为106～107cfu/ml的菌悬液。

试验菌为金黄色葡萄球菌。分组为：①消毒剂+菌液；②(消毒剂+菌液)+中和剂；③中和剂+菌液；④(消毒剂+中和剂)+菌液；⑤正常菌对照；⑥未接种菌的培养基对照。试验结果，当第6组不长菌，第3、4、5组组间菌数差不超过10%，第1组不长菌或菌数远少于第2组，第2组长菌数≥100cfu/ml，可判为所选中和剂及其浓度适宜。
1.3定量杀菌试验
试验采用悬液法。将菌悬液0.5ml与消毒液(对照组用磷酸盐缓冲液)4.5ml混匀。作用至预定时间，取0.5ml菌药混合液，加入盛有4.5ml中和剂的试管中，混匀。中和作用10min，作活菌计数，计算杀灭率。
观察有机物对杀菌效果影响时，向菌悬液中加10%小牛血清。
1.4稳定性试验

将包装完好的商品，随机取样，置于37℃温箱中3个月。于放置前、后，分别取样液检测对金黄色葡萄球菌的杀灭效果。
1.5皮肤消毒试验
随机选60人。左手用消毒液浸泡消毒，右手不作任何处理为对照。采样时，五指并拢，将3cm×3cm规格板放于手指掌面，用浸有中和剂溶液的棉拭用力在方格内反复擦拭，并不断转换棉拭擦拭面。将采样后棉拭头剪断于盛有5ml中和剂溶液的试管中。振打试管以洗脱棉拭上细菌。取洗脱液进行活菌计数，计算灭除率。
2 结果
2.1中和剂选择试验
经对不同复方中和剂比较，以用含0.5%卵磷脂、3.0%吐温80与0.1%组氨酸的磷酸盐缓冲液为中和剂作用较好。该中和剂对滴露的1∶100稀释液(试验用最高浓度)的中和作用可达合格要求(表1)。

2.2 杀菌效果
以滴露1∶400的稀释液对大肠杆菌作用5min，对金黄色葡萄球菌作用10min，杀灭率均达99.9%(表2)。

2.3有机物影响
向金黄色葡萄球菌悬液中加10%小牛血清，以滴露1∶200～1∶100的稀释液对其作用1～10min，与不加小牛血清者相比，杀菌效果略有下降(表3)。

2.4稳定性

2.5皮肤消毒效果
用滴露1∶100稀释液对手浸泡2min。经检测60人，其手上自然菌的灭除率均>90%，平均为98.52%。
3结语
结果表明，以滴露1∶400的稀释液对大肠杆菌作用5min，对金黄色葡萄球菌作用10min，杀灭率达99.9%。以其1∶100稀释液泡手2min，可使皮肤表面自然菌平均减少98.52%。10%小牛血清对其杀灭金黄色葡萄球菌效果略有影响。于37℃存放3个月，该消毒剂的杀菌效果无明显变化。
