    西北药学杂志000301摘 要:采用高效液相色谱法测定大鼠体内8-氯腺苷及其代谢物8-氯肌苷和8-氯腺嘌呤的浓度。试样用乙腈沉淀蛋白后,水层进样,采用YWG-C18。柱,以甲醇-1%醋酸(含0.1%四丁基溴化铵)为流动相,检测波长263nm。8-氯腺苷及其代谢物分离良好,最低检测浓度8-氯腺苷、8-氯肌苷和8-氯腺嘌呤分别为0.10μg/ml,0.50μg/ml,0.25μg/ml,日内精密度和日间精密度均在2.3%~16.0%,三种化合物回收率为70%~99%。

    Determination of 8-chloro-adenosine and its metabolites in rats by HPLC

    Abstract:To determine the concentration of 8-chloro-adenosine and its metabolites in rats with HPLC.The analytical column(250mm×4.6mm)was packed with 10μm YWG-C18H37.The mobile phase was a mixture of methanol and l%acetic acid(containing 0.1%tetrabutyl ammonium bromide)(20:80,V/V).Detection was performed at 263nm.The 8-chloro-adenosine and its two metabolites were separated very well.The minimum detectable concentrations of 8-chloro-adenosine, 8-chloro-adenine and 8-chloro-inosine were 0.1μg/ml,0.25μg/ml and 0.50μg/ml respectively.The precisions within-day and day to day ranged from 2.3%to 16.0%.The mean recoveries ranged from 70% to 99%.
