TNF—α通过内质网应激信号通路诱导肝癌细胞自噬并促进增殖的研究(4)
大量研究已证实,内质网应激与自噬有着密切的关系,内质网应激激活并发生未折叠蛋白反应将引起细胞内自噬水平的上调[1112]。eIF2α作为内质网应激信号通路中的重要信号分子,参与了细胞凋亡、增殖以及炎症调节,其蛋白的磷酸化水平(peIF2α)是提示内质网应激通路激活的标志物之一。在本研究中,我们发现使用TNFα刺激肝癌细胞后,细胞中peIF2α蛋白水平明显升高,提示TNFα诱导的炎症反应激活了HepG2细胞的内质网应激信号通路。
综上所述,TNFα可能通过激活内质网应激通路诱导肝癌细胞自噬而促进细胞增殖。该研究为肝癌细胞自噬的发生机制提供了新的依据,为通过调节自噬抑制肝癌的发生、发展提供科学依据。
