探讨建立大鼠半乳糖性白内障模型的方法(1)
【摘要】目的探讨建立大鼠半乳糖性白内障模型的方法。 方法48只大鼠,随机分为8组,每组6只,双眼均为观察眼,采用腹腔注射法建立大鼠半乳糖性白内障。每日对每只大鼠进行称量,A组:50%D半乳糖10 g/(kg·d);B组:50%D半乳糖15 g/(kg·d);C组:50% D半乳糖10 g/kg,每日2次;D组:50% D半乳糖15 g/kg,每日2次;E、F、G、H的注射剂量等同对应A、B、C、D组,A、B、C、D组在第10日开始加喝10% D半乳糖水。每3 d用裂隙灯观察晶状体的浑浊程度一次。结果观察至第30日时,除最低剂量的E组(3333%)外,各实验组的造模成功率均为100%,A~H各组的死亡率(%)分别为0、3333、6667、3333、0、1667、0、1667。总死亡率为2083%,造模成功率为7083%。结论腹腔注射半乳糖可以成功建立白内障动物模型,且造模的成功率较高,死亡率较低,是较稳定的半乳糖白内障动物模型。

糖尿病患者在2013年約达382亿,2035年预计达到592亿[1]。白内障是糖尿病的最常见并发症之一,更值得重视的是,白内障已被确定为全球视力障碍和失明的主要原因。对于临床研究与治疗,积极寻找病因是治疗及预防疾病最关键的步骤,因此建立稳定可靠的动物模型进行实验研究,有助于日后降低视力损害及白内障盲的发病率[2]。半乳糖诱导的白内障大鼠模型已广泛应用,是较好反映人类糖尿病性白内障和年龄相关性白内障的动物模型。半乳糖模型的建模方法一直存在争议,国外学者认为腹腔注射不能够形成糖性白内障[3]。但是在该领域,我国早期的一些学者发现腹腔注射半乳糖可诱发白内障的形成,还有学者采用球后注射、皮下注射等方法诱发白内障,造模的方式、方法与剂量都不相同,且引起的动物死亡率较高,基本达半数以上[4]。我们经过一系列的实验摸索,探讨半乳糖的用量及方法,旨在提供稳定可靠且死亡率较低的白内障动物模型制备方法,为日后白内障的实验研究及治疗提供一个参考方案。, 百拇医药(钟蕾 王婷 王涛 黎韦华 凌士奇)
