衰老对小鼠小肠上皮形态和增殖速度的影响(1)
【摘要】 目的 研究小鼠在衰老過程中小肠黏膜形态、细胞数目、增殖速度的变化。方法 小鼠按衰老过程分为3组:青年组(3个月组)、中年组(12个月组)、老年组(19个月组),每组9只。每只小鼠按100 mg/kg腹腔注射5-溴脱氧尿嘧啶核苷(Brdu)溶液,并在注射后的24、48、72 h每组各处死小鼠3只,收集空肠和回肠黏膜组织进行形态学研究以及Brdu免疫荧光染色分析细胞增殖情况,并检测增殖相关蛋白β-catenin和cyclinD1的表达。结果 中年组小鼠的绒毛长度、隐窝长度和杯状细胞、潘氏细胞、干细胞数目高于青年组小鼠,而老年组小鼠回肠的杯状细胞、潘氏细胞以及空肠和回肠的干细胞均比中年组降低。Brdu染色显示老年组小鼠Brdu阳性数目和迁移距离比另外2组下降(P均< 0.017)。β-catenin和cyclinD1的表达在老年组小鼠肠黏膜中明显减少(P均< 0.017)。结论 小鼠衰老过程中小肠黏膜细胞增殖能力下降,增殖减慢,隐窝长度减少,杯状细胞、潘氏细胞和干细胞数目减少。

【Abstract】 Objective To observe the changes of the morphology, cell count and the proliferation rate of the small intestinal epithelium in mice during aging. Methods Mice were divided into the young (3-month group, n=9), the middle-aged (12-month group, n=9) and the old groups (19-month group, n=9). Each mouse received intraperitoneal injection of 5-Bromo-2-deoxyUridine (Brdu) at a dose of 100 mg/kg body weight. At 24-, 48- and 72-h after administration, 3 mice were sacrificed in each group and the jejunum and ileum tissues were collected and prepared for morphological observation. The proliferation rate of cells was analyzed by Brdu immunofluorescent staining. The expression levels of β-catenin and cyclinD1 were quantitatively measured. Results Compared with 3-month-old mice, 12-month-old mice had longer villus, deeper crypt, more goblet cells, Paneth cells and stem cells. In the 19-month-old mice, the number of goblet cells and Paneth cells in the ileum, and the number of stem cells in the ileum and jejunum were significantly less compared with those in the 12-month-old mice. Brdu staining demonstrated that the number of Brdu positive cells and the migration length in the 19-month-old mice were significantly reduced compared with those in the other two groups(all P < 0.017). The expression levels of β-catenin and cyclinD1 were significantly down-regulated in the 19-month-old mice(all P < 0.017).
